Target Information and Further Details
PD-L1, encoded by the CD274 gene on chromosome 9p24, is a type I transmembrane protein and a major ligand for PD-1. It is inducibly expressed on immune cells and constitutively expressed on many tumour cells. PD-L1 binding to PD-1 suppresses T-cell activity, fostering immune escape.
Overexpression of PD-L1 is observed in lung cancer, melanoma, renal cell carcinoma, and many other cancers, where it correlates with aggressive disease biology. PD-L1 inhibitors (e.g., atezolizumab, durvalumab, avelumab) are widely used to block the PD-1/PD-L1 axis, reactivating antitumour immunity.
