Target Information and Further Details
HER2 is a receptor tyrosine kinase belonging to the epidermal growth factor receptor (EGFR) family, encoded by the ERBB2 gene located on chromosome 17q12. Unlike other EGFR family members, HER2 does not have a known ligand and functions primarily through heterodimerisation with other ErbB receptors, activating downstream signalling pathways such as MAPK, PI3K/AKT, and JAK/STAT, which promote cell proliferation and survival. HER2 is amplified or overexpressed in approximately 15–20% of breast cancers, as well as in subsets of gastric, esophageal, ovarian, and bladder cancers. HER2 overexpression is associated with aggressive tumour biology and poor prognosis in the absence of targeted therapy. The development of HER2-targeted treatments such as trastuzumab, pertuzumab, trastuzumab emtansine (T-DM1), and trastuzumab deruxtecan (T-DXd) has significantly improved outcomes in HER2-positive cancers. HER2 is also being explored as a target in other modalities, including bispecific antibodies, CAR-T cells, and vaccines.
