Target Information and Further Details
GPRC5D is an orphan G protein-coupled receptor (GPCR) of the Class C family, encoded by the GPRC5D gene located on chromosome 12p13. Unlike other Class C GPCRs, GPRC5D lacks a known endogenous ligand, and its downstream signalling pathways remain poorly defined. Physiologically, it is expressed at low levels in keratinized tissues such as hair, nails, and epidermis, with limited expression elsewhere, making it a relatively tissue-restricted receptor.
In multiple myeloma, GPRC5D is highly expressed on malignant plasma cells, while largely absent from most normal tissues, providing an attractive therapeutic window. It has become a validated target for novel immunotherapies, including bispecific antibodies such as talquetamab (GPRC5D × CD3) and CAR-T cell therapies, both of which have shown potent anti-tumour activity. GPRC5D is also being investigated in other modalities, positioning it as an important emerging target in plasma cell malignancies.
