Target Information and Further Details
FOLR1 encodes folate receptor alpha, a glycosylphosphatidylinositol (GPI)-anchored protein located on chromosome 11q13. It binds and internalizes folates via endocytosis, playing a role in folate transport. Normal expression is limited to specific epithelial tissues such as the kidney, lung, and placenta.
In cancer, FOLR1 is highly expressed in ovarian, endometrial, and some lung adenocarcinomas, where it supports tumour growth and survival. Its tumour-restricted expression pattern makes it an appealing target. Therapies under development include antibody-drug conjugates (e.g., mirvetuximab soravtansine), bispecific antibodies, and CAR-T cells, which exploit FOLR1’s high surface expression for selective tumour killing.
