Target Information and Further Details
EpCAM is a type I transmembrane glycoprotein encoded by the EPCAM gene, located on chromosome 2p21. It functions primarily in cell-cell adhesion, proliferation, differentiation, and intracellular signalling. While normally expressed in many epithelial tissues, EpCAM is upregulated in a wide range of epithelial-derived tumours, including colorectal, breast, ovarian, prostate, pancreatic, and lung cancers. Overexpression of EpCAM is often associated with cancer stem cell properties, increased proliferation, and poor prognosis. Its widespread and high expression in tumour cells, coupled with relatively low expression in non-proliferating normal epithelia, makes it an attractive target for immunotherapy. EpCAM has been targeted using monoclonal antibodies (e.g., catumaxomab), bispecific T-cell engagers, and CAR-T cell therapies. Additionally, it is widely used as a marker for circulating tumour cells (CTCs) in liquid biopsy platforms. Despite its therapeutic potential, targeting EpCAM requires caution due to its expression in some normal tissues, which may lead to on-target/off-tumour toxicity.
