Target Information and Further Details
DLL3 is an inhibitory ligand in the Notch signalling pathway, encoded by the DLL3 gene. Unlike other Notch ligands, DLL3 does not activate Notch receptors but instead functions to inhibit the pathway intracellularly. It is minimally expressed in normal adult tissues but is highly upregulated in high-grade neuroendocrine tumours, particularly small cell lung cancer (SCLC). Due to its tumour-selective expression, DLL3 has been explored as a target for ADCs, including rovalpituzumab tesirine (Rova-T), and is under investigation for CAR-T therapies and bispecific antibodies.
