Target Information and Further Details
Claudin-3 is a member of the claudin family of tight junction proteins, encoded by the CLDN3 gene. It plays a vital role in maintaining epithelial cell polarity and barrier function by regulating paracellular transport. In normal tissues, CLDN3 expression is typically restricted to specific epithelial surfaces. However, it is frequently overexpressed in various cancers, particularly ovarian, prostate, breast, pancreatic, and lung adenocarcinomas. This overexpression is often associated with loss of tight junction integrity and increased tumour invasiveness. CLDN3 is located on chromosome 7q11.23, a region frequently amplified in cancers. Due to its cell surface localization and elevated expression in tumours, CLDN3 has been explored as a target for antibody-based therapies, including monoclonal antibodies and antibody-drug conjugates (ADCs), as well as Clostridium perfringens enterotoxin (CPE)-based approaches, which exploit the ability of the enterotoxin to bind claudin family members and induce tumour cell lysis.
